Toll–Like Receptor 4 Inhibitor TAK-242 Augments Acetylcholine-Induced Relaxation in Superior Mesenteric Arteries of the Streptozotocin-Induced Diabetic Rat
Although vascular dysfunction is a key event in the development of diabetic complications, and abnormal pulse like receptor 4 (TLR4) may contribute to the pathophysiology of vascular disease, a direct link between TLR4 and function of blood vessels in the arterial diabetes remains poorly understood. Thus, to investigate whether pharmacological blockade of TLR4 affect blood vessel function in the superior mesenteric artery (SMA) of streptozotocin (STZ) -induced diabetic rats, SMA isolated from male Wistar rats were injected once with STZ (65 mg / kg, 27 – 34 weeks) treated with TAK-242 (10-6 M), TLR4 inhibitor, for about 1 d using organ culture techniques.
After incubation, functional and biochemical studies performed. In functional studies, treatment with TAK-242 increased by acetylcholine (ACh) induced relaxation of SMA diabetes in the intact condition. Sodium nitroprusside (SNP) induced relaxation was also increased in the group of TAK-242-treated compared to vehicle-treated group. Under the cyclooxygenase (COX) blockade by indomethacin (10-5 M), Ach-induced relaxation was similar in-vehicle group and TAK-242-treated.
Additionally, ACh-induced relaxation in the presence of a combination of nitric oxide (NO) synthase inhibitor, NG-nitro-L-arginine (L-NNA) (10-4 M) and indomethacin (10-5 M) were similar in the group vehicle-and TAK-242-treated. Productions of thromboxane (TX) B2 in the culture medium in the presence of ACh (10-5 M) was lower in the group of TAK-242-treated compared to vehicle-treated group.
These data show that the TAK-242 could improve endothelium-dependent relaxation by partially suppress TXA2 vasoconstrictor or increase NO signal. TLR4 inhibition may be a new therapeutic strategy to assist in the management of vascular complications associated with diabetes.

This study aimed to investigate the effect of box-1-high mobility group (HMGB-1) on the mechanical pain and hypersensitivity underlying mechanisms. Rat primary astrocytes were isolated and treated as specified. A CCK-8 assay was used to determine cytotoxicity and test gap junctional communication is done. Ethidium bromide (EtBr) uptake is used to evaluate the activity hemichannel primary astrocytes.
A rat model of neuropathic pain developed and foot withdrawal thresholds used to evaluate the sensitivity of its hind legs. RT-qPCR and Western blot was used to determine the mRNA and protein expression of the gene, respectively. ELISA was used to measure the release of inflammatory cytokines. Treatment with HMGB-1 increased the expression of both pulses like receptor-4 (TLR-4) and connexin 43 (Cx43) in rat primary astrocytes. HMGB-1 also promoted gap junctional communication between cells and function hemichannel.

Our results also indicate that HMGB-1 is set through the JNK signaling pathway Cx43, and Cx43 are involved in inflammation-mediated HMGB-1 in astrocytes. In vivo analysis supports the idea that HMGB-1-induced mechanical hypersensitivity associated with Cx43. Therefore, we conclude that HMGB-1-induced mechanical hypersensitivity pain occurs through modulation of astrocytic Cx43 through the TLR-4 / JNK signaling.
